It appears that Kancho (aka sticking one's finger/s up anothers arse) is something kids in Japan do as a game. They even have arcade machines, I'm told, that offer people the possibility to do so. This appears to be some bizarre comedy show showing one of these stalls that has a guy inside it instead (so as opposed to quite sanitarily attacking a machine's fake arse, you're getting a bit sticky), so the guy in blue takes out his revenge.

As for the "Sennen Goroshi", it translates, I believe, as "a thousand years of pain". Reference to Naruto episode 4.
